Sao Paulo, Brazil to Prague is an approximately 18.5-hour flight. You can also do a combination of bus and flight. The time zone changes from Brasilia Time (BRT) to Central European Time (CET), which is usually a 4 hour difference. Expect cooler temperatures when traveling from Sao Paulo in April; daily highs in Prague reach 17°C and lows reach 5°C. Wrap up your sightseeing on the 27th (Sat) to allow time to drive to Dresden.
